Definition
Leptin resistance is the blunted hypothalamic response to leptin seen in most cases of established obesity. Leptin crosses the blood-brain barrier less effectively, and downstream signalling is suppressed by chronic inflammation. The clinical consequence: high leptin levels don't curb appetite as intended. Weight loss itself is the most reliable restorer of sensitivity; GLP-1 receptor agonists assist by reducing adiposity.
